It works best under alkaline pH environments and has the physiological role of dephosphorylating compounds. It is available as an opaque white liquid as a suspension in ammonium sulphate. This enzyme is found in both prokaryotes and eukaryotes, with varying structural forms. In humans, it plays an essential role in metabolism within the liver and development within the skeleton. Alkaline phosphatase is used for the conjugation to antibodies and other proteins for ELISA, Western blotting, and histochemical detection. It may be used for protein labelling when high sensitivity is required. It is applied in the removal of phosphate monoesters to prevent self-ligation, which is undesirable during plasmid DNA cloning. Since undifferentiated pluripotent stem cells have elevated levels of alkaline phosphatase on their cell membrane, therefore alkaline phosphatase staining can be used to detect these cells and to test pluripotency.
