Rapamycin by MP Biomedicals is a high grade reagent. Also known by the name Sirolimus, this is a macrolide compound which has immunosuppressant properties in humans. This reagent is available as a white powder which is soluble in ether, chloroform, acetone, DMF, methanol or DMSO, is very sparingly soluble in hexane and petroleum ether, however it is insoluble in water. Rapamycin inhibits the activation of T cells and B cells by reducing the production of interleukin-2 (IL-2). This compound is unique in its ability to inhibit lymphokine induced cell proliferation at the G1 and S phase, as well as an irreversible cellular arrest at the G1 phase in S. cerevisiae cells. It also exhibits selective signal blocking which leads to the activation of p70/85 S6 kinase, which is potentially due to the inhibition of FRAP auto-phosphorylation or protein kinase activity. Angiogenesis inhibition is also exhibited, possibly due to the inhibition of the Akt pathway. Rapamycin also induces autophagy in yeast and mammalian cell lines. These compounds are suitable for research applications.
